QBlueTM Cell Viability Assay

Description
This homogeneous assay involves simply adding a single reagent, the
QBlue reagent to the cell culture and measuring the fluorescence
intensity (excitation wavelength = 530 - 570nm, emission wavelength =
590 - 620nm) after an incubation step. The QBlue reagent, like other
resazurin-based assays, utilises the redox dye resazurin, which is not
fluorescent, but upon reduction by metabolically active cells is
converted into a highly fluorescent product (resorufin). Living cells
can readily reduce this non-toxic reagent and the resulting increase
in fluorescence intensity can be conveniently monitored using a
fluorescence spectrophotometer or plate reader. Non-viable cells have
no metabolic capacity and, thus, will not reduce the dye. Therefore,
the fluorescence intensity observed in this assay is a true measure of
the viable cells. The Qblue reagent has been optimised for maximum
sensitivity, reproducibility and long shelf life. The reagent is
compatible with all culture media and with all liquid handling systems
for high-throughput screening applications in 96-well and 384-well
plates.

Description
The QMTT assay kit provides a convenient, sensitive, quantitative and
reliable assay for determining the number of viable cells in a given
culture. This homogeneous colourimetric assay is based on the
conversion of a tetrazolium salt MTT, a pale yellow substrate, to
formazan, a purple dye. The intensity of the product color, measured
at 550 - 620nm, is directly proportional to the number of living cells
in the culture. Reagents in the kit have been carefully formulated and
optimised for sensitivity, assay robustness and automation.

Description
WST-1 Cell Proliferation Assay Kit provides a safe and simple method
to measure cell proliferation and cell viability. This colourimetric
assay is based on the reduction of tetrazolium (WST-1) salts to
coloured formazan compounds by living (metabolically active) cells.
Tetrazolium salt (WST-1) is cleaved to soluble Formazan dye by
mitochondria dehydrogenase, which is only active in viable cells. The
increase of enzyme activity leads to an increase of the production of
Formazan dye, so the quantity of Formazan dye measured by 450nm
absorbance is directly proportional to the living cells in culture.
The procedure is rapid, accurate and can assay as low as 1000
cells/well
(assay range is 1000 to 100,000 cells /well).

Effect of cell number on absorbance
at 450nm measured using WST-1 Cell Proliferation
Assay Kit Various numbers of K562 cells were added
to the wells of a 96-well plate in 100ml of DMEM
supplemented with 10% FBS. The medium was allowed to
equilibrate at 37°C, 5%CO2 for 30 min. 10 ml of 1:1
mixture of WST-1 solution and Electron Coupling
Solution was added to the wells and allowed to
incubate for 2 hrs at 37°C, 5% CO2 atmosphere. The
absorbance at 450nm was measured using an ELISA
plate reader (reference wavelength 620nm). Each
point represents the mean ± SD of 4 replicates. The
correlation coefficient of the line was 0.996. |

Description
The Malachite Green Phosphate Assay Kit is based on quantification of
the green complex formed between Malachite
Green, molybdate and free orthophosphate. The rapid colour formation
from the reaction can be conveniently measured on a spectrophotometer
(600 - 660nm) or on a plate reader. The assay is simple and fast,
involving a single addition step for phosphate determination.

Malachite Green Phosphate Assay, kinetics of the
chromogenic reaction.
80µL of standard phosphate solution
was transferred to a clear-bottom 96-well plate.
20µL of the Malachite Green Reagent was added and OD
at 650 nm was read every minute. The chromogenic
reaction was completed within 10 minutes. The signal
was stable between 10 and 20 minutes.
Storage conditions
The Malachite Green Reagent and standard is stable
for one year when stored at 4°C.
