pNPP Phosphatase Assay Kit

Description

Para-nitrophenyl phosphate (pNPP) is a chromogenic substrate for most phosphatases such as alkaline phosphatases, acid phosphatases, protein tyrosine phosphatases and serine/threonine phosphatases. The reaction yields paranitrophenol, which becomes an intense yellow soluble product under alkaline conditions and can be conveniently measured at 405nm on a spectrophotometer. This homogeneous “mix-andmeasure” assay involves simply adding a single reagent to the phosphatase, and measuring the product formation using any absorbance reader. The assay can be conveniently performed in cuvettes, tubes or multi-well plates at either room temperature or 37°C. The kit reagents have been optimised for long shelf life and maximum reproducibility. The reagents are compatible with all liquid handling systems, and bulk reagents are available for high-throughput screening of phosphatase inhibitors.

Storage conditions

The reagent should be stored in the amber tube at -20°C. The Assay Buffer and Stop Solution are stable at 4°C and room temperature, respectively. Shelf life: 12 months.

 PBlue Phosphate Assay Kit

Description

The PBlue Phosphate Assay Kit is based on an innovative formulation of the malachite green dye. The PBlue reagent forms a blue coloured complex with free orthophosphate. The rapid colour formation from the reaction can be conveniently measured on a spectrophotometer (600 - 660nm) or on a plate reader. This single reagent “mix-and-measure” colourimetric assay has been optimised to offer superior sensitivity and prolonged shelf life. The assay is simple and fast (quantitation of free phosphate within 30 minutes), involving a single addition step for phosphate determination. Applications include phosphatase assays, lipase assays, nucleoside triphosphatase assays and high-throughput screening for phosphatase inhibitors (drug discovery).

• Stable reagent – no precipitation occurs and no filtration of reagent is required

• High sensitivity and wide detection range – 20pmoles of phosphate (useful range between 0.4µM and 50µM phosphate)

Storage conditions
